Why is HT2 exposure greater?
Two possible reasons broadly. It was expressed to a greater degree or is more accessible to the antibody.
Why did the tagging reduce titres?
Optimisation needed.
Why did HT1 have such a negative effect?
- Interfered with activation
- Interfered with fusion
- Altered membrane trafficking.
ANOVA and Dunnet’s
The data means were assumed to follow a normal distribution. ANOVA avoids cumulative type 1 errors.
Dunnet’s chosen as it compares each mean to a control mean.
Tukey compares all means to each other.
Would any other statistical tests have been appropriate?
No
Why no positive control for the Ig staining?
Couldn’t find one.
Cumulative effect of HT1/3 but not HT2/3. Care to hypothesise why?
The expression of HT2 could have been great enough that the tag sequestered available antibody.
