Blocks potassium channels
- inhibits depolarisation and hence prolongs the action potential
Limiting factors of amiodarone
Half life of 20-100 days (loading doses often used)
Should be given into central veins (causes thrombophlebitis)
Has proarrhythmic effects
P450 inhibitor so decreases metabolism of warfarin
Adverse effects of amiodarone
Thyroid dysfunction : hypo and hyper
Corneal deposits
Pulmonary fibrosis
Liver fibrosis
Peripheral neuropathy
Photosensitivity
Slate grey appearance

Why is metformin contraindicated in recent MI
Because metformin can cause lactic acidosis if taken during a period where there is tissue hypoxia eg recent MI, sepsis, AKI, severe dehydration

Inhibits sodium reabsorption by blocking the Na+-Cl symporter at the beginning of the DCT
