Western Half Life function
-Determines the half life of a specific protein under specific conditions
Step 1
Treat cells as desired
What happens right after treatment?
After treatment, the cells will make proteins, some at varying levels
Step 2
-After treatment, add puromycin or cyclohexamide to stop any additional translation
Step 3
Extract total protein from cells at various times
Step 4
Protein concentrations
Step 5
Add loading buffer of
- SDS and beta-ME
- Glycerol
- Bromophenol blue
Step 6
Boil samples
Step 7
Run samples on an SDS Page gel
Step 8
Transfer to a nitrocellulose membrane
Step 9
-Block membrane with BSA or milk
Step 10
-Add primary Ab to protein of interest and wash
Step 11
-Add secondary Ab which is conjugated to HRP and wash
Step 12
-Add chemiluminescent reagent that interacts with HRP, giving off light
Step 13
-Expose membrane to film so we can now see the protein levels
