Light microscopy
-view entire cell (morphology)
-less magnifying power
- view living cells
Simple Light Microscope
- single lens to reflect natural light to the sample slide
Compound light microscope
-uses two or more lens to reflect light off the sample slide making the specimen look bigger
Dissecting light microscope
- single objective lens, two eyepiece lens
-used for dissection and 3D specimens
Method in light microscopy:
Standard Light Microscopy (brightfield)
Light passes directly through the
specimen. Has a lesser quality of observation
due to unstained samples having lesser
contrast.
Method in light microscopy:
Phase Contrast Microscopy
-manipulated by condenser inducing contrast
- better quality but creates halos
-dark parts= denser
-used for observing living unstained cells
Method in light microscopy:
Differential Interference Contrast Microscopy (Normanski)
-similar to phase contrast but has plane-polarized light, Normanski prisms
-internal structures
Modified prism of made of two merged crystalline quartzes
Wollaston prism
Method in light microscopy:
Fluorescence microscopy
-stained samples
-sample appears bioluminescent
-fluorescent signifies cellular protein or organelle
