Plasmid Flashcards
1
Q
What is RNA 1?
A
- A negative regulator
2
Q
What is RNA 1’s function in ColE1?
A
- Binds with RNA 2 to stop further replication occurring.
3
Q
How is RNA 1 regulated?
A
- Its concentration is proportional to the copy number of the cell (More plasmid = more RNA 1)
4
Q
What is the complex called when RNA 1 and RNA 2 bind?
A
- Kissing complex
5
Q
What binds RNA 1 and RNA 2?
A
- Rom and Rop protein
6
Q
What occurs after RNA 2 binds to origin of rep?
A
- RNA 2 DNA hybrid allows RNA 2 processing, RNase H binds, polymerase binds, Primer maturation, Replication
7
Q
What is plasmid incompatibility?
A
- Two plasmids with the same origin of replication have the same system for copy number. The regulatory system cant tell them apart, therefore one is lost to chance.
8
Q
How do you avoid plasmid incompatibility?
A
- Make sure the plasmids have different origin of rep and Ab resistance markers so can differentiate between them.
