What is the mechanism of action of chloroquine
A
1.Inhibit parasite from hemoglobin digestion——-> decreasing nutrient amino acids for the parasite
2.Inhibit parasite heme polymerase——-> accumulation of toxic heme

what are the therapeutic uses of chloroquine
A
1.Prophylaxis and treatment of chloroquine sensitive malaria ( followed by primaquine to eradicate dormant form—-> prevent relapse)
- Amoebic hepatitis and amoebic liver abscess
- Rheumatoid arthritis (anti-inflammatory effect)

Adverse effects of chloroquine
A
- Headache
- C.V.S: hypotension and arrhythmia (quinidine like action)
- GIT: Nausea, vomiting and diarrhea
- Eye: corneal opacity——> blurred vision and retinopathy
- Hemolytic anemia: in G6PD deficiency

Quinine is
A
- Absorbed well after oral administration
2.interfere with heme polymerization——> death of RBCs
3.used in chloroquine resistance falciparum
4.weak muscle relaxant
5. not used as prophylactic (too toxic)

Adverse effects of quinine
A
- Quinidine like reaction——>hypotension and arrhythmia
- Eye: blurred vision and blindness
- cinchonism: tinnitus, headache, dizziness & visual disturbance
- Black water fever and hemolysis
- Oxytocic on uterus——> abortion
