MOA: Activation of descending serotonergic inhibitory pathways in the CNS
Clearance: Hepatic and renal
Consideration: potentiates opioid therapy, do not use with infant with G6PD, prophylactic administration prior to vaccination may decrease immune response.

A

MOA: Inhibits COX-1 and 2 enzymes, which results in decreased formation of prostaglandin precursors.
Clearance: Hepatic metabolism, renal excretion

MOA: Binds with stereospecific receptors at many sites within CNS increases pain threshold, alters pain reception, inhibits ascending pathways. Highly lipophilic, redistributed into muscle and fat
