How to plot a log concentration response curve

A
  1. IF conc. is in nM, convert to M.
  2. Log Molar concentrations.
  3. Plot the scale on the Y-axis (response[mm]).
  4. Plot the scale on the X-axis (Log[Conc]).
  5. Plot line of best fit (sigmoidal line)
  6. Find the half of the Y-axis maxinum and trace down its correlated X-axis value on the graph.
  7. Reverse log this value.
  8. EC50 should be in M

A
  1. Determine Bmax Y-value
  2. Half the value
  3. Trace the Y-value to the line to determine the correlated X-axis value.
  4. The X-value will be the estimated kd .

A

A value which equates to the affinity of a binding interaction. The affinity is also determined by the dissociation constant (KA). A lower KA equates to a higher affinity

  • Low kd = HIGH AFFINITY
  • High kd = LOW AFIINITY
