Pharmacogenetics
A field of research that studies how a person’s genes affect how he or she responds to medications
Pharmacogenomics
Deals w/ simultaneous impact of multiple mutations in the genome that may determine the patient’ response to drug therapy
Pharmco genetics/genomics will allow:
More rapid determination of stable therapy dose
Better prediction of dose than clinical method alone
Use of medications not generally considered
What may affect genetic influences on Drug Response?
Uptake of medication
Drug receptors
-Decreased uptake due to receptor differences and can lead to toxicity
-Intracellular breakdown of drug metabolites can reduce efficacy of the drug
What are reasons a person would react differently to a drug based on DNA?
Having receptor recognize drug

Drug Breakdown
Drug breakdown in Body:
Drugs are considered foreign by the body which works to excrete the substance (liver, kidney, intestinal cells)
Hepatic enzyme system works to inactivate drugs and facilitate excretion
Role of CYP Enzymes:
Cytochrome P450 enzymes are essential for metabolism of many drugs
May be metabolized by one or multiple enzymes
Any difference in availability of enzyme or efficacy of action will change how the drug metabolizes
