PCR Flashcards
What PCR?
A biochemical process capable of amplifying a single DNA molecule into millions of copies in a short time
What are the three steps of PCR?
Denaturation
Annealing
Extension
Describe the denaturation stage of PCR?
dsDNA templates heated to separate the strands
Describe the annealing phase of PCR
Short DNA molecules (primers) bind to flanking regions of the target DNA
Describe the extension phase of PCR
DNA polymerase extends the 3’ end of each primer along the template strands
At which temperature is PCR performed?
Stage 1 - 95
Stage 2 - 61
Stage 3 - 72
What are the clinical applications of PCR?
Fingerprinting
Genetic diagnosis
Viral detection
What is in the mastermix?
dNTP
MgCl
Reaction buffer
Taq polymerase
