(Non-cancer) Genetic Disorders Flashcards
What is the average age of onset of Huntington’s disease?
30-50yrs
What does HD cause?
Progressive chorea, dementia and psychiatric symptoms
What inheritance characteristics does HD show?
AD with genetic anticipation
What does myotonic dystrophy cause?
Progressive muscle weakness and myotonia (failure to relax) in early adulthood
What inheritance characteristics does DM show?
AD with genetic anticipation
What inheritance characteristics does cystic fibrosis show?
AR
What does CF cause?
Increased thickness of secretions - results in recurrent lung infections and exocrine pancreatic insufficiency
What does newborn screening for CF measure in blood?
Immunoreactive trypsin (IRT) level
What is inheritance pattern of neurofibromatosis 1 (NF1)?
AD
What are some minor manifestations of NF1?
Cafe au lait macules and neurofibromas
Short stature
Macrocephaly
Lisch nodules on sclera
What are some serious manifestations of NF1?
Scoliosis Tibial fractures Phaeochromocytomas Sarcomas Optic pathway gliomas
What is inheritance pattern of DMD/BMD?
X-linked recessive
Which protein is faulty in DMD/BMD?
Dystrophin - normally forms link between F-actin (intracellularly) and the dystroglycan complex in muscle fibres
What will boys with DMD have massively increased levels of in their blood from birth?
Serum creatinine kinase - CK leaks out of damaged muscle fibres into serum
Which type of deletion is seen in DMD?
“out-of-frame” deletion - results in earlier age of onset than BMD
