Alzheimers Disease and Genetics
Familial form of Alzheimer’s – early onset
5% of families - mutated form of amyloid precursor protein (APP) production of more β amyloid
Non-familial genetic risk factor – APOE – E4 allele
E4 allele – 15% of general population
60% of Alzheimer’s patients
One SNP determines difference between ApoE3 and ApoE4
(change an amino acid in protein, alternate structure)
Polygenetic factors: Twin Studies
Monozygotic Twins (genetically identical). 100% Huntington's. 50% Schizophrenia and 69% Bipolar disorder. Dyzygotic twins: same sex (50% genetics). 50% Huntington's, 15% schizophrenia, 13% bipolar disorder.
Inbred Strains (Mice)
Inbred strains will have different genetics, neurobiology and behaviour. The study was about mice drinking alcohol (ethanol 10%) C57 will drink more with the sucrose inside it but the BDA’s wont drink that much more than usual
Knockout and Transgenic Mice
Transgenic (changing a section of DNA) - when you think that a certain gene is causing something so you take that and manipulate that gene into other mouses genomes in other mice
