It synthesize prostaglandins and thromboxanes from arachadonic acid
How do NSAIDs interact with Cyclooxygenase?
they down regulate its activity
What kind of receptors are prostaglandin receptors?
They are g-coupled receptors
What are the Clinical Presentations of osteoartheritis?
• Deep, aching pain and pain on motion
• Stiffness in affected joints
• Pain resolves with motion, recurs with rest
• Duration of pain is less than 30 minutes
• Pain related to weather
• Limited joint motion
• Hands: involve distal interphalangeal joints, Herberden nodes, Bouchard’s nodes
• Knee: pain related to climbing stairs, transient joint effusion
• Hips: Groin pain during weight bearing exercises, stiffness, especially after
activity, limited joint movement
• Spine: Lumbar involvement, parasthesisa, loss of reflexes
What is unique about Asprin?
It irreversibly binds to COX enzymes. This means that it has a duration that is much longer then its half life.
What are the Propionic Acid Derivative NSAIDs?
Ibuprofen, Naproxen, Ketoprofen, Flurbiprofen, Fenoprofen
What is unique about Naproxen?
It has a long half life (18 hours) and has the lowest risk for cardiovascular issues of its class
Indomethacin, Sulindac, Etodolac, Diclofenac, Ketorolac are all what kind of drug?
Acetic acid derivative NSAIDs
What is unique about Ketorolac (toradol)
It is used to treat sever pain in patients that cant use opioids. It also has a black box warning that it should not be used for more than 5 to 7 days.

it is not indicated to treat feaver
