Molecular biology and Medicine Flashcards
1
Q
What are the steps in cloning?
A
Digest genome –> separate fragments –> clone –> sequence –> assemble
2
Q
What are the 3 sites in bacteria that we are concerned with?
A
- Polylinker sites
- Antibiotic/ampicillin-resistant gene
- Origin of replication
3
Q
What method is used for cloning when we use bacteria?
A
- Lac operon.
- IPTG is used
- LacZ gene is replaced with the gene of interest
4
Q
What is trimethylaminuria?
A
Bacteria in the gut convert choline to trimethylamine (which smells like fish) – in normal people, this is converted into trimethylamine N-oxide by flavin-containing monooxygenase 3 (FMO3).
However, in people with trimethylaminuria, there is a mutation in FMO3, so trimethylamine accumulates – so it smells like fish.
