What is myelin?
white substance forming sheaths around axons
Made of protein, cerebrosides, cholesterol, phospholipids, etc
What produces myelin?
Schwann cells in the PNS
Oligodendrocytes in the CNS
What part of myelin is left after processing?
Neuroketain, a proteolipid
How does myelin stain in an H&E?
Stronger pink than non-myelinated nerves
Difficult to pick out
What staining method is often used to detect myelin?
Immunohistochemical techniques
What is myelin demonstration useful for?
Demonstration of demyelinating disease like multiple sclerosis
What are the positive and negative techniques used in staining for myelin?
Positive - stains degenerate myelin
Negative - stains normal myelin
What is an example of a negative technique for myelin?
Luxol fast blue
What is Nissl substance?
Acidic aggregations of rough endoplasmic reticulum
Also called tigroid or chromidal substance
Where is Nissl substance found?
In the cytoplasm and dendrites of neurons
What are good stains for Nissl substance?
Cresyl Echt Violet

What types of dyes are the ones used for Nissl substance?
Basic analine dyes
What happens to Nissl substance when nerve damage occurs?
It starts to disappear, first around the nucleus then throughout the cytoplasm
Describe the 3 stages of myelin degeneration
Early - myelin still present
Intermediate - lipid breakdown to neutral fats
Final - neither fat nor myelin present
Describe what will stain in the 3 stages of myelin degeneration
Early - myelin with Luxol fast blue
Intermediate - neutral lipid with oil red O
Final stages - nothing
