  1. Place 2 drops of donor’s serum in a 5 ml test tube.
  2. Add 1 drop of 3-5% patient’s red cells.
  3. Mix and centrifuge at 3400 rpm for 15 seconds then read
    for the presence of agglutination or hemolysis.

A
  1. Wash with NSS three to four times.
  2. Add two drops of AHG reagent.
  3. Mix and centrifuge for 15 seconds at 3400 rpm.
  4. Read for hemolysis or agglutination with or without microscope.
  5. Add 1 drop of check cells if negative.
