what is the principle of minor crossmatching
A
this detects the presence of antibodies in the donor’s serum that will damage or destroy the patient’s RBCs

A
- 5 ml test tube
- NSS
- Water bath
- AHG reagent
- Centrifuge
- Microscope

specimen used for minor crossmatching
A
a clotted or anticoagulated blood drawn within 72 hours of testing may be used

adequate complement and calcium

immediate spin/saline phase
A
- Place 2 drops of donor’s serum in a 5 ml test tube.
- Add 1 drop of 3-5% patient’s red cells.
- Mix and centrifuge at 3400 rpm for 15 seconds then read
for the presence of agglutination or hemolysis.

protein phase/thermal phase
A
- Add 2 drops of 22% bovine albumin.
- Mix and incubate at 37 deg C for 15-30 minutes.
- Centrifuge for 15 seconds at 3400 rpm then observe for
agglutination or hemolysis.

AHG phase
A
- Wash with NSS three to four times.
- Add two drops of AHG reagent.
- Mix and centrifuge for 15 seconds at 3400 rpm.
- Read for hemolysis or agglutination with or without microscope.
- Add 1 drop of check cells if negative.
