Very thick peptidoglycan layer; stains purple
What are the gram stain steps?
- Crystal violet, 30 sec
- Rinse with DI water
- Gram’s Iodine (mordant): 30 sec
- Rinse with DI water
- Acetone alcohol (decolorizer): rinse off ASAP
- Secondary stain: Safranin, 1 min
- Rinse with DI water
- Blot dry with kimwipe
How do you determine cell size?
Field diameter/#of cells that fit across the field of view
What is the function of a mordant?
Prevents the crystal violet from leaving the gram + cell wall
What step in the gram stain is differential & why?
The decolorizer is the differential step, because gram negative bacteria will lose the purple color.
What are the 3 basic cell shapes?
Bacilli, cocci, spirilla
What are the 4 cell arrangements
Single (any shape)
Diplo (bacilli or cocci)
Staphylo (cocci only)
Strepto (cocci or bacilli)
