Microscopy Flashcards
What is Kohler illumination?
Kohler illumination is used in light microscopy to evenly illuminate a specimen.
How do you use Kohler illumination?
The condenser knob is used to focus the light on the specimen and use the field
diaphragm to control the illuminated area.
When do you use Kohler illumination?
It is used when taking precise measurements or taking pictures of a specimen.
What does parfocal mean?
Lens that stay in focus when you change the magnification. Allows you to shift
between magnification without the need to refocus.
What are dissection microscopes used for?
Dissecting microscopes are used for larger, 3D specimens (insects, plants. etc.).
What kind of magnification and light sources do dissection microscopes have?
Possesses low magnification, usually between 5x-50x magnification. Also
possesses two light sources (one at the top is reflective and one at the bottom is
transmitted.
What are compound microscopes used for
Compound microscopes are used to magnify small, microscopic specimens on a
flat plane (cells, bacteria, etc.).
What magnification do compound microscopes have?
Provides high magnification, from 40x-1000x.
6 basic rules when handling microscopes
Only adjust coarse focus at the lowest magnification
-
Carry with two hands, one at the base and one at the arm.
-
Use proper cleaning tissue when cleaning lenses.
-
Don’t drag microscopes across a surface. Lift to move.
-
Secure slides using the clips to ensure stabilization
