microscopy Flashcards
What are the 5 types of light microcopy?
bright field, phase contrast, differential interference, dark field, and fluorescence
what is bright - field microscopy?
Bright field microscopy is the simplest type: in order to generate contrast, cell components are stained/ dyed to make them visible
what is phase shift microscopy?
Two types: in phase and out of phase –> when light moves in phase, light intensity is high. when light moves out of phase, peaks and troughs are added but cancels out, generating a dim image
what is differential interference microscopy?
Differential interference microscopy manipulates phase differences to visualize edges or boundaries
what is dark-field microscopy?
the only light that goes to our eye are the ones being scattered by the cell. You cannot see the outside of the cell as the image appears bright against a dark background
What is immunocytochemisrty?
a lab technique to indicate where a certain protein is using the body’s own antibodies
What is transmission Electron Microscopy?
In TEM, electrons shoot out of the electron gun at high velocity, focusing on the specimen, generating the image (higher electrons, shorter wavelength). Images that use TEM typically has a high resolution to see smaller and specific parts of the subject
What is scanning electron microscopy?
SEM uses heavy metals on the specimen and scattered electrons bouncing off the it to generate an image. SEM produces a textured, 3D image but uses lower resolution and sees only external features
