Pros of fluorescent microscopy
rapid, relatively inexpensive.
Cons of fluorescent microscopy
wide variation in results, not a sole diagnostic indicator, not used for monitoring sites for activity bursts.
What does a healthy site look like?
- 24-48 hours old.
- scanty and cells well dispersed
- mainly coccoid, some rods
- no motility
- desquamated epithelial cells
- neutrophils may be visible.
What bacteria are normally present in a healthy site?
Actinomyoces, Streptococci.
What does a gingivitis site look like?
Dense cells, periphery with monolayer, cocci, short rods, filamentous cells, clusters of motile rods.
Give an example of filamentous bacterial cells
F. nucleatum.
What does an active periodontitis site look like?
Mainly motile plaque.
Similar to gingivitis but higher % of motile and spirochaetes.
Epithelial cell debris.
Neutrophils may be observable and degranulating.
Describe microscopy method sampling.
- Wet plaque sample from pocket with sterile curette.
- Mix with BacLight reagent.
- Live bacteria = green. Sytox Green.
- Dead bacteria = dull red. Propidium Iodide.
