microscopes Flashcards
Advantage of a light microscope
can observe living cells for example mitosis
What are the 2 disadvantages of a light microscope?
Stain will kill living cells
Low resolution (meaning cant see all the living organelles in the cell)
Magnification and resolution
x1500, 200nm
Why cant a light microscope have a high resolution?
Because it has a short wavelength of visible light (400nm - 700nm)
Does it pass through the specimen?
Yes
What is a electron microscope?
Uses a beam of electrons and needs to be in a vacuum
What are the two types of electron microscopes?
SEM (Scanning electron microscope) and TEN(Transmission electron microscope)
Why cant a electron microscope be used on living cells?
Because they would die in vacuum as there is no particles in there
Do electron microscopes pass through the specimen?
Yes some parts of the specimen more easily than others
Advantages of electron microscope (2)
High resolution
Have 2000x than light microscope
Disadvantage of electron microscope
Uses a vacuum so cany us e living cells and require very careful staining and the specimen has to very thin
Define resolution
The minimum distance between 2 objects where they can be seen as two separate objects
Describe TEN
Uses beam of electrons
Electrons pass through the specimen and has to be very thin
Ultra structures are seen
Black and white images produced
Higher resolution
Magnification 100,00x - 500,000x
Resolution 0.1nm
Describe SEM
Uses beam of electrons
Electrons pass back and forth across the specimen
Magnification=50,000x
Resolution 0.1nm
Define staining
Provides a contrast between organelles and cytoplasm allowing structures to be observed
