What is Trypsinization?
Digestion of protein by trypsin
What is peptide map fingerprints?
peptides unique for a protein
How does mass spectrometer separate ions?
mass/charge ratio
Mass constant, different charges
Single charged, doubly charged, triple charged
The degree of deflection depends on what?
Molecular mass of ions (m/z ratio)
Smaller gets deflected more (requires less force)
Bigger gets deflected less
What is ESI-MS?
knocks out electrons making protein positively charged
Proteins with the most charge reaches detector first while least charge will arive last (most + AA which are Cys, Thr, Arg)
What is MALDI-MS?
treat with acid which will give protons
all +1 charge with different masses
Since all have +1 charge, the m/z ratio will start from the smallest to the largest
What is the tallest peak called?
base peak
What is the rightmost peak called?
parent peak
