Making and purifying proteins Flashcards
Step 1
- Retransform bacteria with the engineered plasmids
Plate on LB/AMP (Luria-Bertani/Ampicillin) plate where bacteria make protein of interest
Step 2
Grow up lots of bacteria
Transfer to 1L flasks
Step 3
Lyse bacteria (Kill/Explode bacteria) (Sonication)
Collect lysate
Step 4
Centrifuge to Separate bacterial debris from proteins
Collect proteins
Step 5
Purify the wanted antigen away from other proteins
Column Chromatography
Separate proteins by size
Works like a filter
Step 6
We now have our desired antigens from a specific virus, which we can inject into the body (known as Subunit Vaccine).
The immune system then can react to all the separate antigens the same way it would with an actual virus, and create an immune defense against that virus, without having the body contract the disease.
Somewhat similar to Inactivated Virus Vaccines
