Light Microscopy Flashcards
Common types of microscopy
Bright field
Bright field:uses optics and white light to contrast objects and backround
FLuoresence
Localises tagged proteins on cells
Live microscopy
Allows study of cells movement
Confocal microscopy
Higher resolution
Digital microscope
Acquired by computer
Electron microscopy
Electrons as a source of investigating
Illumination radiation
Reaches 1nm size objects, used in nanotechnology & biology
Magnification
Is the enlargement of the appearance of an object
Contrast
Describes an ability to detect an object against a backround.
Resolution
A term that describes our ability to distinguish discrete objects
Bring two objects brought closer and closer together until they no longer appear to be seperate objects
Contrast
Refers as the difference in light intensity between the specimen image and the adjacent backround relative to the overall backround intensity.in general contrast is measured by the relationship between the highest and lowest intensity
Resolution
Ability to distinguish (separate) 2 objects situated close enough
When to use immersion oil, 1
Only be used if you have an immersion oil lens.
Lens will have HI (homogenous immersion)
Immersion oil is best for using samples that are dead or are not moving and no thicker than a few micrometers.
Very small moving protists can occasionally be viewed with IM
When to use immersion oil 2
Should be used anytime you want to view a clearer image at 1000x
-striatitions in skeletal muscle
Disadvantages to using immersion oil ?
Wet mount slide must be incredibly secure in order to use immersion
Is switching between dry 40x and 100x make sure not to get oil on the 40x lens which could damage it
