How does a cell differentiate into TH1?
PAMPs from intracellular bacteria and viruses, cause
dendritic cells and macrophages to make IL12
and nk cells to make IFN-y
How does a cell differentiate into TH2?
caused by IL4
How does a cell differentiate into TH17?
PAMPs from extracellular bacteria and fungi cause
dendritic cells to make IL1, IL6, and IL23 (TGFb is also impt)
What is the effect of Th1
To activate macrophages by classical pathway. increase Ros/NO, increase IL12 (more Th1), increase MHC, increase TNF (recruit lymphocytes)
What do CD8+ Tcells do (aka CTLs) ?
they use perforin (to make a pore in the cell) and then inject granzymes (to induce cell to undergo apoptosis)
What is the effect of Th2?
they mainly fight helminthes? promote eosinophil recruitment (IL-5) IgE production (IL-4) alternative pathway of macrophage activation (IL-4 and IL-13)
what is the effect of Th17?
IL17 and IL22 recruit neutrophils and defensins(epitherlial tissue). They mainly deal with extracellular pathogens
The role of each Th1,2,17 in disease is?
Th1= autoimmune, chronic inflammation Th2= allergies Th17= autoimmunity
