Lecture 6 - Cell Biology & Protein Purification Flashcards

1
Q

What are the features of the peptidoglycan layer?

A
  • The PG layer is mechanically tough.
  • It is held together by covalent bonds.
  • Must be artificially damaged to release proteins from the cell.
  • Most bacteria require an intact PG layer to survive.
  • Also a target for antibiotics.

Q

What are some of the components needed to partially reproduce ‘physiological’ conditions?

A

Almost all “buffers”
* Buffer molecule (phosphate, Tris)
* Salt (NaCl, KCI)

Popular additives
* Other salts (MgC12, CaC12)
* Detergents
* Protease inhibitors
* Nuclease inhibitors
