DNA strands are unwounded by helicase to create the replication fork. DNA pol III (replicase) synthesizes the leading strand continuously from an RNA primer, whereas lagging strand is synthesized in okazaki fragments using a new RNA primer for each fragment. RNase removes the primers and DNA pol I uses nick translation to fill in the gaps.
