Lecture 26 Flashcards

1
Q

Describe the lifecycle states of Caulobacter crescentus

A
  1. Swarmer cells - swims in aquatic environment and find a surface to attach to. Undergoes development where it produces a ‘hold fast’ (sticks itself to a surface). At this point it CANNOT DIVIDE
  2. Stalked cell - cell post attachment, only time it can divide

Q

Describe Illumina Sequencing in terms of Transposon location

A

It gives the transposon and a small piece of surrounding DNA allowing them to know where the Tn jumped in

Q

What is the effect of a Tn jumping into a high fitness ORF?

A

These aren’t essential but are good for bacterial growth, a Tn insertion will cause bacteria to grow slower

Q

Where might essential genes be able to tolerate inesertions and why is this?

A
  • At the far end, majority of the protein has been formed and is functional at this point
  • Might tolerate right at the beginning
