Describe the lifecycle states of Caulobacter crescentus
A
- Swarmer cells - swims in aquatic environment and find a surface to attach to. Undergoes development where it produces a ‘hold fast’ (sticks itself to a surface). At this point it CANNOT DIVIDE
- Stalked cell - cell post attachment, only time it can divide

Describe Illumina Sequencing in terms of Transposon location
A
It gives the transposon and a small piece of surrounding DNA allowing them to know where the Tn jumped in

What is the effect of a Tn jumping into an essential gene?
A
No growth

What is the effect of a Tn jumping into a high fitness ORF?
A
These aren’t essential but are good for bacterial growth, a Tn insertion will cause bacteria to grow slower

Where might essential genes be able to tolerate inesertions and why is this?
A
- At the far end, majority of the protein has been formed and is functional at this point
- Might tolerate right at the beginning
