mRNA produces what type of library?
cDNA library
A genomic library is made from which molecule?
DNA
What is a cDNA library?
Contains all mRNA in an organism (from genes being expressed in that cell).
How do you construct a cDNA library?
- isolate mRNA (oligo - dT column)
- Anneal oligo - dT primer and copy the DNA using reverse transcriptase.
- Treat with RNA seH (attacks RNA of RNA/DNA hybrid strands) -> left with only DNA.
- Add DNA Polymerase I to make dsDNA
- Clone into plasmid or vector.
You can’t amplify with PCR unless….
… The sequence is already known (so you can then design primers).
Which library has all genes in an organism?
BAC library.
The BAC genomic library includes what in it?
Introns, promoters, repeat sequences, transposable elements, etc.
Plasmid cDNA libraries have what?
Coding regions and adjacent transcribed sequences only.
Which library has a lower complexity?
cDNA library.
How big are the inserts in cDNA?
1-2kb on average (plasmid or phage vectors)
How many genes per clone does the BAC genomic library hold?
Multiple (or partial) genes per clone.
What method do you use to find a gene with hybridisation?
DNA probe. Use a probe from another species with 70% bp homology (using PCR). Finding a sequence with high Met and Trp is good as there is only one sequence for this. “Degenerate probe”.
What method is used to find a gene detecting an expressed protein?
Using antibodies. Need to purify and inject into the animal, then make an expression library.
Which method is used to clone conserved core functions?
Complementation of E.coli/yeast mutants.
