Lecture 13 - Herpes 3 Flashcards
What are the 4 phases in the cascade of herpes virus gene expression?
1) immediate early phase
2) early phase
3) DNA replication phase
4) Late phase
The cascade of gene expression from early to late phases involves what kind of activation?
Transactivation
What occurs during Herpesvirus immediate early phase?
Synthesis of transcription factors that govern the sequential synthesis of all later proteins
What occurs during herpesvirus early phase?
Synthesis of its viral DNA-dependent DNA polymerase and other replication machinery
What occurs after herpesvirus early phase?
DNA replication phase with viral DNA-dependent DNA polymerase
What occurs during herpesvirus late phase?
Synthesis of structural proteins?
what does phosphonoacetic acid do and what is it used for?
Inhibits DNA polymerase activity and can be used experimentally to separate early events from late events
What are concatemers?
long continuous DNA molecule that contains several copies of the viral genome
What creates herpes virus concatemers?
The viral DNA polymerase during DNA synthesis
Why are concatemers made?
In order to allow packaging of the herpesviral genome into the nucleocapsid
How are individual genomes removed from the concatemer for packaging?
Terminal sequences at the end of each genome facilitate cleavage
What protein do herpesviruses use to compete for host translation machinery? What kind of protein is it and what does it do?
VHS, it is a viral endonuclease that targets host mRNAs and degrades them
