Trypsin cuts the C terminal end of Lys and Arg
Chymotrypsin cuts c terminal end of Trp, Phe and Tyr (aromatic groups)

- the reaction rate in increased by reducing the activation energy, more products are made so delta G is a negative number

Enzyme saturation tests, xray crystallography, change in spectroscopy.

A
Vo is initial rate, Vmax is the highest rate, Km is half of Vmax, Kcat is turnover # (# of substrate molecules converted into product units are 1/S), 1/kcat is time required for one molecule to be processed.
