Lecture 10 Flashcards
Which factors lead to growth in quiescent cells?
E2F, myc, cyclin E
Which 2 genes does E2F activate that lead to p53 activation?
ATM and ARF
What are the 4 p53 pro-apoptotic cofactors?
JMY, ASPP1, SPP2, DINP1
What does p53 activate that leads to growth arrest?
P21
Myc works as a heterodimer with
Max
When Max binds mad, what’s the effect?
Repression of mitogenic signals, growth inhibition
What is MAD?
a repressor of myc - binds max instead of myc,
c-myc
overexpression in many tumors, translocation in burkytt’s lymphoma
n-myc
amplification in neuroblastoma
l-myc
amplification and rearrangement in lung cancer
Can myc alone transform cells? Why?
No, because of the fail safe mechanism and bc myc and many myc targets require post translational modifications
What does overexpression of beta catenin cause?
Cancer
What are beta-catenin’s jobs in the cell?
Cell adhesion and transcription regulation
What does Beta Catenin activate when it goes tot he nucleus?
Cyclin D and myc, which cause proliferation
What, other than proliferation, can beta-catenin activate?
It can transcriptionally activate Arf –> activate p53 –> apoptosis
