Lab 1 - Microscopy Flashcards
The course adjustment knob should only be used with which objectives?
4x (scanning) and 10x (low power)
What is the function of the iris diaphragm?
It is located under the stage, and may be opened or closed to the desired size, thereby regulating the amount of the light by increasing or decreasing the diameter of the shaft of light entering the microscope
What is the condenser?
A group of lenses that cause light rays to converge on the surface of the microscope slide.
Why must you use oil when looking through the 100x lens?
You have to use oil because the slide is so magnified that the air between the lens and the slide can distort the image. When you use oil it creates a seal between the lens and the slide, so the image is not warped by the air.
When you look through the microscope, is your image represented as the specimen as it as mounted on the slide or is inverted?
It is inverted.
What is a prepared slide?
A slide that has been manufactured for repeated use. To make one, cells are placed on a slide, stained to make them visible under the microscope, and sealed permanently under a tiny piece of glass called a coverslip.
One micrometer is equal to _____ millimeters.
1000
Which objective should always be in place when beginning to focus?
4x
Which objective has the largest diameter of field
4x
Which objective has the smallest diameter of field
100x
If you are using the 4x objective which focusing knob[s] can you use?
Course focus & fine focus
If you are using the 40x or 100x objective which focusing knob[s] can you use?
Fine focus
What is always used for cleaning the microscope lens
Lens paper, lens cleaner
What is always used for cleaning blank or specimen slides?
Lens paper, lens cleaner
How do you calculate total magnification?
magnification of objective lens x magnification of eyepiece
