Possible treatments for RTK overactivation

A

Monoclonal antibodies - prevent ligand binding and/or dimerisation

TKIs (tyrosine kinase inhibitors) - prevent RTK activity

Why are MAbs ineffective against ligands in a majority of tumours?

A
  • Most tumours are polygenic and not caused by exclusively one pathway
  • Other ligands are around and may ‘take over’ the job of that suppressed ligands
