Possible treatments for RTK overactivation
A
Monoclonal antibodies - prevent ligand binding and/or dimerisation
TKIs (tyrosine kinase inhibitors) - prevent RTK activity

Why are MAbs ineffective against ligands in a majority of tumours?
A
- Most tumours are polygenic and not caused by exclusively one pathway
- Other ligands are around and may ‘take over’ the job of that suppressed ligands

Why are downstream treatments less effective than upstream ones?
A
Hard to stop other branches form overcompensating - shows step-wise model of multiple pathways
