Journal 1 - Wnt-2 and Lung Cancer Flashcards
Wnt-2 Signaling
A molecular pathway where the Wnt-2 protein binds to Frizzled receptors, triggering a series of cellular events that influence cell growth and development
Wnt-2
A protein that binds to Frizzled receptors to activate the Wnt signaling pathway, playing a key role in cell development and cancer progression
Frizzled-8 (FZD8)
A receptor protein on the cell surface that binds to Wnt proteins, initiating the Wnt signaling pathway which is important for cell development and growth
Beta-Catenin (β-catenin)
A protein involved in cell signaling and gene transcription, playing a crucial role in the Wnt signaling pathway and cell-cell adhesion
Endogenous
Originating or produced within an organism, tissue, or cell.
Exogenous
Originating or produced outside an organism, tissue, or cell.
Constitutive Expression
Continuous expression of a gene, not regulated by external stimuli.
TCF
T-cell factor, a transcription factor activated downstream of Wnt signaling, regulating target gene expression.
Reporter Assay
An assay that measures the activity of a gene’s promoter, often used to study signal transduction pathways.
RT-PCR (Reverse Transcription-PCR)
A technique to amplify and detect specific RNA sequences, used to study gene expression.

WNT Canonical Pathway
- Wnt binds to frizzled
- Wnt signaling is activated
- DVL helps stabilize B-Catenin
- B-catenin enters cell nucleus
- B-catenin associates with (LEF/TCF) transcription factors
- Promotes transcription of genes shown to cause cancer.
Summary of Figure 1A
Wnt-2 is overexpressed in cancer, but what receptor is it expressed with? When tested in 293T cells to see which frizzled receptor could bind to Wnt-2 (and activate TCF factor) Frizzled-8 had the highest effective interaction with Wnt-2.
fzd-8 and wnt-2 activity super high together
Q: What is the relationship between Wnt-2 interactions with it’s receptors (frizzled) in lung cancer
What method was used in Figure 1A?
TCF Reporter Assay - it detected activation of Wnt canonical pathway
assay that measures the activity of a gene’s promoter
