JC3: Signalling, receptors, and pharmacology (3) Flashcards
1
Q
Describe the stimulation of adenylyl cyclase
A
- An agonist binds to the GPCR
- There is GTP-for-GDP exchange which results in an active αs-GTP complex
- This causes an increase in enzymatic activity
- ATP is converted to cyclic AMP which activates cyclic AMP-dependent protein kinase (PKA) and adenylyl cyclase
2
Q
Describe the inhibition of adenylyl cyclase.
A
- An agonist binds to the GPCR
- There is GTP-for-GDP exchange which results in an active αi-GTP complex
- This causes an decrease in enzymatic activity
- ATP is not converted to cyclic AMP so cyclic AMP-dependent protein kinase (PKA) and adenylyl cyclase are not activated
3
Q
How does cyclic AMP exert the majority of its actions?
A
Through cyclic AMP-dependent protein kinase (PKA)
