Infectious Causes Of Cancer Flashcards
Major mechanism for KSHV transformation is
The viral protein Latency Associated Nuclear Antigen 1 (LANA 1)
LANA 1 binds
P53
KSHV also plays a role in
Immune evasion & Angiogenesis
EBV mediated Transformation
EBNA 2 & LMP 1(latent membrane protein)
EBNA 2
Acts as a transcriptional activator for many downstream genes. Unregulates B cell antigens CD21 (binds EBV and activates B cells) and CD23(growth signals)
Alter expression of host oncogene cMYC
LMP 1
Believed to act as a constitutive member of TNF reception superfamily. Resemblance to CD40 so mimic B cell activation B cells proliferate with no stop signal creating Anti apoptotic & cytokine genes
Tumors consistently have chromosomal translocation
in the region c-myc proto-oncogene (8q24)
Hit & Run hypothesis
Viral infection was needed to begin the process of transforming but translocation events in the host cell are necessary factors for maintaining oncogenic status
HTLV 1 causes
Adult T cell leukemia/lymphoma
Acute transforming
Retrovirus carriers an oncogene which is then incorporated into the host genome under viral promoter
Slow transforming
Dwells on location of viral genome is inserted into host genome eg HTLV typically inserts 10kb of TF binding sites
