Impregnation and Embedding Flashcards
Process whereby the clearing agent is completely removed from the tissue and is replaced by a medium that will completely fill all the tissue cavities
IMPREGNATION or INFILTRATION
• Process by which the impregnated tissue is placed into a precisely arranged position in a mold containing a medium which is ten allowed to solidify
EMBEDDING
Embedding is also known as
CASTING or BLOCKING
Medium used in impregnation will be the same for embedding
TRUE
• Simplest, most common and best embedding medium used for routine tissue processing
PARAFFIN WAX
• Defined as obtaining a continuous ribbon of sections from a paraffin block and placing all the sections on multiple slides
Serial Sectioning
How long may Tissue blocks and unstained mounted tissue sections may be stored in paraffin after impregnation without tissue distortion
INDEFINITE PERIOD
• Prolonged impregnation will cause
excessive tissue shrinkage and hardening
• Inadequate impregnation will cause
retention of clearing agent
becomes soft and shrunken
blocks crumble or break up when floated in a water bath
• Paraffin processing is recommended for fatty tissues
FALSE
medium should be in liquid state
Infiltration
medium should solidify
Embedding
microtome used
Use sledge or sliding microtome
melting point of commonly used paraffin
56 degree Celsius
recommended temperature for manual infiltration
2-5 degree Celsius above the melting point of the wax
