Immunoassays Flashcards
What is an immunoassay?
a test to detect and quantify an antigen in your sample
What do antibodies recognize?
Antibodies recognize antigens through specific epitopes
Monoclonal
One type of Ab
Recognizes one single epitope
Polyclonal
Mixture of Abs bind to the same Ag
Multiple epitopes recongized
How can we detect primary Abs?
-Option A: directly bound to fluorescent dyes, compounds, enzymes for detection
-Option B: Primary antibody + secondary antibody that is conjugated with a detection tag
Direct methos can avoid ____ and cau be used with
avoid cross-reactivity between 2o, can we use multiple abs from same species
Types of detection tags:
Fluorescent tags
Enzyme-linked Ab
Sometimes the antigen is in very low quantity in the sample… thus
we need to modify our assay to make it more sensitive signal amplification
How can we amplify the signal?
Multiple 2nd Abs can bind to each 1 Ab
Immunohistochemistry
Selectively imaging antigens inside cells of tissues
Flow cytometry
Detect and measure characteristics of cell pops
ELISA
High throughput method of quantitatively measuring protein concentration compared to standard
Immunoblotting
Western blot - detecting specific proteins
Southern blot
detecting specific DNA seqs
Northern blot
Detecting specific RNA seqs
