How can a T cell receptor be designed to recognize a specific antigen?
An Immunoglobulin gene generated to recogenize a apecific antigen can have the intracytoplasmic portion modified to activate the same signalling pathways as a natural T cell receptor. Such chimeric T cell receptors are protent stimulators of both CD4 and CD8 T cells.
How can :CD8+ T cells be genetically modified to be more or less potent?
Crisper gene modification using single guide RNA (sgR:NA) lenti viral infection with CAS9 electroporation (SLICE) of a ensitized CD8 T cell can rank genes in terms of the diffenence between knockout and native gene function when responding to an antigenic stimulant. Removing suppressing genes and replacing defective activating genes can markedly increase cytotoxicity.
