How does diazoborine inhibit ENR?
-diazoborine sits on top of nicotinamide ring, blocking substrate entry
-boron is covalently bound to NADH ribose
What are the problems with using diazoborine as an inhibitor for ENR?
-v. easy for bact to become resistant (simple changes like Gly to Ala)
-B is also v. toxic to us
How does triclosan inhibit ENR?
-mimics enolate anion intermediate
-stacking interactions with nicotinamde ring of NADH
-upper ring interacts with where patethneine arm of ACP would be, blocking substrate entry
Why is triclosan a more suitable inhibitor for ENR than diazoborine in?
-not toxic to us (diazoborine is)
-harder for bact to develop resistance to triclosan
-v. high affinity
