how to investigate effect of pH on the rate of an enzyme controlled reaction Flashcards
1.
prepare a range of different pH solutions
2.
mix each pH solution with enzyme e.g. amylase
3.
prepare test tubes also of starch solution
4.
equilibrate the tubes of pH solution of amylase and starch in a water bath to 37 degrees
5.
mix together the pH amylase solution and starch
6.
every 30 seconds remove a sample
7.
place sample on a dimple tile containing a drop of iodine - note the colour change
8.
continue until the iodine is no longer BLACK - showing that STARCH has been DIGESTED - record the time taken to DIGEST the STARCH
9.
repeat for each pH and at least 3 x for each pH
10.
calculate a mean time for each pH
11.
control variables - temperature, concentration of starch, amylase
