Major cause of juvenile hemochromatosis (AR).

HJV acts as a coreceptor for BMP receptors – specifically, its action as a coreceptor for BMP6 controls hepcidin metabolism.

Pathogenic variants most common in families of Greek descent (G320V founder mutation).
