First 2 phases of Bacterial Growth?
Lag: No macroscopic increase in cell population. Attempt to minimize loss. (To minimize use large inoculum and medium)
Exponential: Occurs when cell population is NOT limited. Characterized by doubling time.
Last 2 phases of bacterial growth?
Stationary: Number of bacteria being produced = # of bacteria dying (dynamic condition)
Death: Cell lysis, nutrients run out, cells present but not viable
How is cell number monitored?
Microscopic count
Viable cell count (petri dish)
How is cell massed measured?
Cell dry weight (centrifugation)
Turbidity (optical density)
3 types of inhibition?
Competitive: Inhibitors and substrates are competing for the same active site
Uncompetitive: No affinity to enzyme itself but bind to the complex reducing effectiveness.
Noncompetitive: Bind on sites other than active site and reduce enzyme affinity to the substance.
Fermentation Patterns?
Type l: Product is associated with cell growth (overall yields = instantaneous yield)
Type ll: Instantaneous ≠ overall
Type lll: non growth associated (secondary metabolites)
