How does the cas9 nuclease function.
The Cas9 nuclease together with a specific guide RNA locates the target DNA sequence that requires editing. Once in place, it make a precise double stranded cut. Once cut, the cellular material recognises the cut and gathers the machinery to repair the double stranded break. This is done through non-homologous end joining or homology- directed repair.
How has cas9 been change for base editing.
Modified Cas9 protein makes single strand break and deaminase enzyme converts one base to another.
How has cas9 been change for prime editing.
Modified Cas9 protein makes single strand break and with reverse transcriptase, can copy new sequences into nicked stand based on an extension of the guide RNA.
