1) Denaturation: DNA sample is heated at 94 degrees C
2) Annealing: primers bind to DNA and polymerase attaches and starts copying DNA at 50-64 degrees C
3) Extension: at 72 degrees, optimum temperature for polymerase and extension of fragment
