What is the proper working concentration for vespene? How SHOULD it be made?
1:128 is the proper working concentration.
If you expect to add liquids to the waste container, make a final concentration of 1:100 to ensure no matter what volume the waste reaches, it will be enough to inactivate.
(10mL vephene in 1L water).
How long does vephene last for? What is the contact time?
2 weeks.
10 minutes
Do you have to wash hair in the shower?
Only if working with HPAIV or SARS-CoV-2
What do you do if you are locked out of the shower room?
- During normal business hours, call integrated security. Provide the door number, wait some time, then try to prox in.
- During off-hours call Pitt police and ask to page the BST3 operating engineer. They will come and unlock the door.
- If in an emergency, use the glass break. Pitt police and the BSO must be notified.
What are some general principles of working in a BSC at BSL3?
- Check the thimble monitor and the magnahelic gauge prior to work.
- Ensure front and rear grilles are clear
- Keep aerosol generating equipment at the back of the hood.
- When finished work, bag up solid wastes. Spray bag down with vesphene, place into a second bag, and vesphene that off prior to removal from the hood.
- For liquid wastes, ensure everything is in contact with vespene for 10 minutes prior to removal, and then dump down the drain.
What would you do if your outer gloves become contaminated?
Dispose of the gloves and replace with a new pair.
What if both layers of gloves become compromised and you were potentially exposed to a Select Agent?
- Remove both glove layers and wash hands.
- Don two new pairs of gloves.
- If skin was exposed, secure SA materials and exit following normal procedures.
- Contact supervisor and EHS to report potential exposure.
What is the door policy in the RBL?
Never hold or prop a door open as it will trigger and alarm and can potentially compromise directional airflow in the facility.
If doors are needed open longer than 30 seconds, notify BSO prior to event.
Describe the proper spatial and temporal separation of SAs.
SAs that require spatial and temporal separation include RVFV and HPAIV.
In addition to full decon of BSC between agents, a 20-30 minute time period must be observed when using different agents in the same BSC.
Describe how you would operate the autoclave?
- Bring bagged waste to autoclave.
- Check current cycle to make sure it is empty.
- Open the autoclave door and place materials inside.
- Run on cycle p17!!!
- Log the waste contents
If in the CC:
1. Look at the ticket to ensure the cycle is complete.
2. Open the autoclave door and place bagged waste on the rack.
3. Go to log book and enter info such as date, time, what the materials are, and if sterilization was achieved (autoclave tape).
4. Place ticket in the envelope.
Describe proper traffic through the ABSL3 suite.
- NEVER use the soiled corridor to move between animal suites.
- When leaving one room with animals, spray off boots and change outer gloves.
- Place equipment in the airlock prior to entry. When on hot side, ensure nobody is in the airlock and retrieve equipment without stepping over threshold.
