Gel Electrophoresis Flashcards
Why cut DNA?
Isolating the restriction enzymes are use to manipulate DNA
Blunt ends
When both strands of DNA are cut directly opposite each other
Sticky ends
When the second strand is not cut directly opposite the first cut
Seperating DNA
Electrophoresis is used to sort out fragments into groups of their length. Copies are made by PCR (Polyerase Chain Reaction).
Electrophoresis
1) DNA is isolated from cells
2) DNA cut into fragments
3) Fragments loaded onto gel
4) Electric field applied
Electrophoresis electrical charges
DNA fragments have a net negative charge. They move to the positive poll. Shortest fragments move the quickest
