gel electro phoresis Flashcards
What is gel electrophoresis’s use
Gel electrophoresis is a technique used to separate charged molecules like DNA, RNA, and proteins based on
their mass.
what do combs do
Used to make wells in the gel
what does the gel tray do
Gel tray holds the newly poured gel
what does the gel casting chamber do
Chamber where gel is poured and solidified
what does the buffer chamber do
The buffer chamber holds the gel casting chamber and the buffer
what is the polymerase chain reaction
PCR is a technique that amplifies or
makes multiple copies of a short strand of DNA or genes
what are the stages of the DNA polymerase chain reaction cycle
Denaturing ,Annealing, and extending
what happens to the temperature during the DNA polymerase chain reaction cycle
The temperature starts at 94-95 degrees celcius then it drops 40 degrees celcius then it goes back up to 72 degrees celcius
What is the original copy called
template
what does every cycle do
Every cycle doubles the number of copies exponentially, if you run 30 cycles you will obtain around a billion
copies
what does primer do
primer is like the key, it starts the process
what is the thermal cyclers use
The thermal cycler is used to amplify DNA via the polymerase reaction
what is the centrifuge use
The centrifuge is an instrument that is used
to separate particles in a mixture based on
its density
Rpm=
rotations per minute
what are three precautions for the centrifuge
- Use the right-sized tubes, the tube should match the rotor.
- Make sure you balance the tubes, if you add 5 ml to one tube and 2 ml to another tube the separation will be
affected. All the tubes need to contain the same volume of liquid. - Balance the tubes. If you place two tubes on one side of the rotor, place two more on the opposite side to
balance the rotor. If you have only one tube, fill another tube with the same amount of water and place it on the
opposite side
