Forward Genetics Flashcards
1
Q
Give the definition of forward genetics.
A
Seeks to find the genes encoded by DNA that are responsible for a phenotype of interest.
2
Q
Give the classical genetics strategy.
A
- isolate mutants with a phenotype of interest
- choose a good organism
- find a mutant of that organism – can generate mutations by applying mutagens
- screen for mutants with desired phenotype - define genes responsible for these phenotypes
- clone/identify the genes
- analyse the genes to predict encoded proteins
3
Q
Describe the process of cloning genes by mutant rescue.
A
- transform cells with gene/cDNA library
- isolate transformants which rescue, or restore wild-type phenotype
