Q

Give the classical genetics strategy.

A
  1. isolate mutants with a phenotype of interest
    - choose a good organism
    - find a mutant of that organism – can generate mutations by applying mutagens
    - screen for mutants with desired phenotype
  2. define genes responsible for these phenotypes
  3. clone/identify the genes
  4. analyse the genes to predict encoded proteins

Q

Describe the process of cloning genes by mutant rescue.

A
  • transform cells with gene/cDNA library

- isolate transformants which rescue, or restore wild-type phenotype
